I just got the Strecthmo dlc the other day. You have to play the tutorial puzzles (they start automatically when you open the game) and then the dialog to purchase the DLC will pop up at the end of them.

And you can buy the route DLC in Fates at the title screen I think. You only have to play a couple hours up to the route split to open up the dragon gate on the world map and buy the map DLC. Same thing with Awakening. You have to play a couple hours to open up the dragon gate option on the world map. In Echoes I think you just have to get to the world map, so just like the opening cutscenes and the first battle so maybe about half an hour or so.

I remember seeing an article pointing out that while the Layton vs. Ace Attorney DLC is free, you have to beat the game to access it. There might be some games with Spotpass features too, like I think Devil Survivor 2 had some demons via spotpass it rotated through.
