I like when one or several main themes get variations throughout the soundtrack. That's usually pretty cool, especially when the melody is the same but due to changes in speed or accompanying instruments or such, the tone is completely different.
But should an entire soundtrack be one song remixed a whole bunch? No. Not even for old school short games w/ a very small soundtrack.

Hard to think of video game examples at the moment... Baten Kaitos soundtracks tend to use similar motifs in a bunch of songs, for one...
I usually find it more in anime. Ronin Warriors uses several instrumental variations of each of its three main songs (Samurai Heart, Stardust Eyes, and Faraway) and they're all great. Cutie Honey series always uses a ton of variations of the theme song in the soundtrack to good effect, too.
