there's a number of different hunters you can encounter and fight over the course of the game. rifle spear hunter is probably one of the more difficult ones for me, at least as far as a 1 v 1 fight. basically for the exact reasons you mentioned. I was actually just fighting him again yesterday on a casual run that I'm doing and I was thinking about this as well.

Numbing Mist is the item that blocks healing. good catch on noticing that. I never used it on my first run because I just assumed it wouldn't work on a boss. whoops. your other options are to simply pile on damage while she's casting the heal, since she won't counter attack or dodge during the entire length of the animation. this will usually cause her to only heal like 1/4 of the amount she's trying to heal because your damage offsets her healing by a good amount. or simply kill her so fast that she never gets to the stage where she'll heal during the fight. using Fire Paper will greatly boost your damage output. you should have picked some up along the way.

exploring the side areas that you currently have access to is a great way to find upgrade materials so you can improve your weapon and greatly improve your damage. as a rule of thumb -- explore as much as possible and then fight the boss after you've found all of the items available.

there's also people who you can find and send back to the cathedral ward. make sure you're knocking on the different doors and windows with the lamps burning outside of them!
