Okay, I learned a trick for Jacob & Esau. There's these alternate floors you can go to, and their item rooms have two upgrades in them. Normally, a single character would grab one and the other disappears. But since you can have one brother stand still while the other can still move, that means you can get them in position to grab both items at once, so they can each get almost as many upgrades as a single character.

Still a challenge to get to the end, though. You still have to manage their health so it's fairly even, and a bad room can leave one brother at low health. I think the devs knew they were a bad character, since they have strong item unlocks for getting completion marks with them. One particular one can actually downplay/negate the inherent downsides of the other characters.
