You know, technically Tonks doesn't really do much. And I don't care.
These are both primarily humor/novelty characters. I find Tonks entertaining and funny. I don't find Lockhart funny; he just annoys me. (I kind of like him after he loses his memory, I guess).
And...I find it hard to write detailed analysis of humor/appeal.

Yep, both of these characters start off primarily as comic relief characters- one a hilariously incompetent professor whose narcissistic mannerisms and devilishly handsome looks belie a cunning (if still talentless) man, and the other a clumsy Metamorphmagus who uses funny slang, has to be reminded of her duties, and will screw up her face just to make her friends laugh.
While my initial impression of Lockhart may have been stronger (I found him hilarious and didn't find Tonks' unique antics quite as entertaining), Nymphadora grew on me quite a lot. Enough to become my first/second favorite female character in the series (depending on how I'm feeling about Hermione on a given day). The character herself becomes a lot deeper around the start of the sixth book, where she seems even less focused than usual, distracted and aloof, lacking her usual sense of humor. There's that little Easter egg with her Patronus- a nice touch- and BAM! Romance out of... almost... nowhere. After attempting to mislead me into believing that Tonks was in love with Sirius (something I didn't buy for a second since ew, that was her cousin) it's revealed that she was in love with Remus.
Even though the relationship is only built throughout one book, I can forgive it for two reasons: 1) These two really work together as a couple (for all of one year. :'(). I can definitely see each's negative attributes balanced out by the others' more positive traits and visa versa. And 2) Neither is what I'd call a main character. You pair a huge somebody with a relative nobody and the pairing will cause an uproar. Pair two nobodies together and no one will give a damn.

Gilderoy Lockhart, all day every day! He was always the most entertaining character in the series for me. Tonks gets a few points, but she loses out by having her Black blood tainted with a mudblood father. I can't believe people would choose a nearly useless character over one of the greatest heroic wizards of all time!
Also if we take film representation into account as well, Branagh beats the really odd choice they went with for Nymphadora. No contest there.

(6)Gilderoy Lockhart Tonks was a pretty minor character, and she didn't do anything that really stood out to me. She married Lupin (I think?), but their relationship wasn't that developed, there were just a couple scenes of him not wanting to get involved and then I guess he changed his mind? I don't really remember, didn't make a big impression on me.
Gilderoy on the other hand was pretty funny. Yeah, he was a total idiot, but he also got repeatedly humiliated throughout the book (like when he starts the dueling club), and his ultimate fate in CoS was a thing of beauty. Then they make you feel a little bad for him by showing him in St. Mungos, but you can't say he didn't deserve what he got.
