It seems like there is an appetite recently for starting to localize previously Japan-only games here. Personally I want to see a god damn Front Mission complete collection, but what's at the top of everyone else's list?

I'd be ecstatic for an English translation of Shin Onigashima (Famicom Disc System). The soundtrack is one of the best of its era and the storyline and characters seem charming. Sadly, though, I doubt this would ever happen - translating it would be a huge undertaking for little financial benefit.

I'd also love to see official English versions of Mother 3 and Captain Rainbow, though at least those have been fan-translated.

...actually probably Dai Gyakuten Saiban (the Ace Attorney spin-off we never got) or the Dragon Quest Monsters 1+2 remake. For some reason I was only thinking of old games we never got.

We've been doing soooooooo much better in the last couple of generations about this kind of availability. Even stuff like Dai Gyakuten Saiban got/are getting fan translations at least, if not late releases like Metal Wolf Chaos, PSO2 and Seiken Densetsu 3.

I think at this point for me the big one is Yakuza Ishin, but Fate Extra CCC is right behind it.

I'll throw out a few that come to mind:

-Famicom/Game Boy Wars series: a compilation for the Switch would be dope
-Mother 1, Mother 3: honestly insane that these haven't been localized given Smash Bros
-Final Fantasy III: we've STILL never gotten an official localization of the original game, except on mobile I think
-Treasure of the Rudras: seems like a really neat gimmick
-Terranigma: has this been made available virtually? If not, that's completely insane as it would take zero effort
-Live a Live: I think this is every JRPG fan's #1
-Fire Emblem: there are a few in the series that haven't been remade right?
-Front Mission: pretty criminal that these haven't been packaged up yet
-Langrisser/Growlanser: ditto^
-Policenauts: this will never, ever happen in today's culture
-X-Men vs Street Fighter: the US PSX port of this sucks right? Has the Saturn one ever gotten a virtual release?
-Tales of Phantasia: not the butchered GBA release

The original 2D Final Fantasy 3 has never made it to the West, but the 3D remake came to mobile, DS, and maybe PSP??

Zero no Kiseki and Ao no Kiseki, especially as it becomes increasingly important to the story of the series overall.

Chaos;Head because it remains bizarrely the only Science Adventure VN that isn't localized or announced for localization.

Ciel nosurge in my impossible dream world.

Probably some others too but those were the first to come to mind.
