That means I need to study the bible and recm watch all episodes of Supernatural right? Would a Supernatural campaign work? The players will play as hunters. And it's rather simple. Their goal is to stop Lucifer from destroying earth.

You wouldn't really need to study the Bible though. It might make for some fun quotes, but Supernatural goes by a very loose interpretation.
I imagine it'd work well. D&D starts off by killing random monsters of the week like wendigos and then goes on to killing giant demon gods and shit, just like Supernatural. I think it'd be pretty simple to translate some D&D monsters into Supernatural ones.
