It could be on HBO. In the first episode, the earth is about to be destroyed. Some disaster. Maybe they could keep it mysterious on what happened

Some people are able to get off the planet into space on a ship. It has 500 people on board. According to a Google search, that is the absolute smallest number that could repopulate the species after a massive near extinction event

So it's a big ship. This show would be be set in the future. Maybe like 300 years from now

The last 500 humans set out into space on a trip towards Alpha Centauri in the hope of finding an earthlike planet there

They don't actually know if it exists. Also the trip will take 50 years. They don't have cryogenic. Everyone has to just live and form a society and have kids and wait

During the journey there could be drama like murders on board or rebellions to replace leadership

also maybe the horror aspect could be weird stuff happening in space like people seeing ghost like creatures on board. Maybe it turns out to be aliens from a different plane of existence, seeping into the ship. Something like in the movie Event Horizon

Story spans decades. Every once in a while there is a time skip. Series finale finally reveals if they find a new planet humans can live on

Theres been a few shows and movies on this already. Some similar ones are the 100 and passengers.

Edit: there was also another show that escapes me, only 2 or 3 long episodes. And then a horror movie where they're being hunted on a spaceship on their way to a new world.

500 would only be enough if it was particularly planned out from the start pichu. The number would have to be in the thousands to avoid genetic defects and such if it was completely random subsect of the population

Would be cool if it was a generational epic, seeing the story of the early travellers and then after their arc is done, do a time skip to their grandkids or something.

This is basically Stargate Universe mixed with BSG, which is what SGU was trying to be. They end up on the other side of the universe when they dial the 9th Chevron.

There's also Pandorum, which another user mentioned. They leave Earth on an ark with 60k people because Earth's resources ran out due to overpopulation.

Some of the crew wakes up and encounters Gollum like creatures. The twist is that its been a thousand years since they left and reached their destination 800 years ago, but landed and sunk into the ocean. Others woke up and eventually devolved over the hundreds of years.
