The joke is that Squall, a rather callous communicator (Talk to a wall), has the option to answer the question rudely. Him being able to question if they are even a woman is supposed to be insulting because hes saying they are ugly. Its purely coincidence that the individual is either crossdressing, or in the starting stage of being trans. Hard to know since Japan has things like traps and other tropey gimmicks about this.
