Since all 4 ports can all send simultaneous inputs, you can hook up 2 players. Player 1 uses A and R (and presumably the control stick) and controls the bro in front in the overworld and Mario in battle. Player 2 uses B and L and controls the bro in the back in the overworld and Luigi in battle. It's pretty dope, especially with Bros. Attacks. Superstar Saga also has Gameboy Player compatibility, so they clearly had something like this in mind.
