Yup. Since all 4 ports can all send simultaneous inputs, you can hook up 2 players. Player 1 uses A and R (and presumably the control stick) and controls the bro in front in the overworld and Mario in battle. Player 2 uses B and L and controls the bro in the back in the overworld and Luigi in battle. It's pretty dope, especially with Bros. Attacks.
