Inspired by SNESDrunk's "Unconventional Ways to Play Classic Super Nintendo Games" videos. (You should definitely check SNESDrunk out).
Finding alternate ways of playing a game, whether by doing a self-imposed challenge, or just playing a game differently than it was intended, are a good way of spicing up games you've played hundreds of times.

I'll start with the 30 Melon Challenge in Yoshi's Story, where you try to get all of the 30 Melons in each level, and NO OTHER FRUIT. It's basically like Hard Mode for the game. Particular Mushroom has a video that goes into more detail.

The Nuzlocke Challenge has got to be one of the more famous instances of this. From Bulbapedia:

The Nuzlocke Challenge has only two rules that must be followed: The player may only catch the first wild Pokmon encountered in each area, and none else. If the first wild Pokmon encountered faints or flees, there are no second chances.

When I used to play rocket league, my only goal was blowing other people up. Didn't give a shit about winning, I just wanted to make people go pop and get mad.

Randomizers or Shufflers seem to be pretty popular among the speedrunner community lately, they've been pretty fun to watch

Alternative controllers are pretty neat too, which I guess is technically an alternative way to play games... watching a guy speedrun Alien Isolation with a DJ Deck was fun

As you can probably guess, I don't exactly do alternative methods myself :P

Doom has a few categories of alternate playstyles. One I discovered recently is "Stroller":

Play on Ultra-Violence
Not allowed to harm enemies directly/intentionally
Not allowed to run
Not allowed to strafe

Driving correctly in GTA.
