If you've still got open slots, pick The Kid vs The Guy, from I Wanna Be The Guy. What other rivalry produces so many deaths? What other protagonist goes through so much hassle to make it to his adversary? None! It's the best of both worlds - it functions as a joke nomination if that's your thing due to the general lack of seriousness, but it's also a real hero/villain pair from a fun game. Everyone should want it in.

Sakura Kasugano / Karin Kanzuki: Also from Street Fighter; the female version of Ryu / Ken, imo.

Jin Uzuki / Margulis: from Xenosaga; a pretty great rivalry from a pretty great series. In fact, Shion aside, the stories of the other main characters are pretty interesting.

KOS-MOS / T-elos: Xenosaga; um...hot female androids.

I wouldn't call any of these locks. Dan / Sagat has the best chance of getting in, followed by KOS-MOS / T-elos. The other two are the longest of longshots.
