Seen it before (and apparently there might be other ones like it but they aren't available), including a few re-watches, and it's still oddly entertaining even though the only really funny moments is watching the professor react. Despite the material not being that funny, it had an embarrassing number of breaks from "professional" comedians, especially Jonah Hill who has difficulty getting through his lines. The others are luckier because they can mug a little to hide it.

I genuinely only know depictions like this from American tv and movies. I have been to anime conventions with friends in the past and to me the movie is not a caricature or even an exaggeration of that culture, it is just making fun of stereotypes I am not even sure exist.

It's making fun of weebs (the old school kind; not the new meaning that basically refers to anyone who likes anything Japanese). Weebs are absolutely like this. I've seen quite a few online and a handful in person over the years.

I didn't want those people to be acknowledged by the mainstream because once it happens, some people will apply the stereotype to the whole fanbase. That and I knew it would just be painful to watch.
